    Prediction of the functional class of metal-binding proteins from sequence derived physicochemical properties by support vector machine approach

    Metal-binding proteins play important roles in structural stability, signaling, regulation, transport, immune response, metabolism control, and metal homeostasis. Because of their functional and sequence diversity, it is desirable to explore additional methods for predicting metal-binding proteins irrespective of sequence similarity. This work explores support vector machines (SVM) as such a method. SVM prediction systems were developed by using 53,333 metal-binding and 147,347 non-metal-binding proteins, and evaluated by an independent set of 31,448 metal-binding and 79,051 non-metal-binding proteins. The computed prediction accuracy is 86.3%, 81.6%, 83.5%, 94.0%, 81.2%, 85.4%, 77.6%, 90.4%, 90.9%, 74.9% and 78.1% for calcium-binding, cobalt-binding, copper-binding, iron-binding, magnesium-binding, manganese-binding, nickel-binding, potassium-binding, sodium-binding, zinc-binding, and all metal-binding proteins respectively. The accuracy for the non-member proteins of each class is 88.2%, 99.9%, 98.1%, 91.4%, 87.9%, 94.5%, 99.2%, 99.9%, 99.9%, 98.0%, and 88.0% respectively. Comparable accuracies were obtained by using a different SVM kernel function. Our method predicts 67% of the 87 metal-binding proteins non-homologous to any protein in the Swissprot database and 85.3% of the 333 proteins of known metal-binding domains as metal-binding. These suggest the usefulness of SVM for facilitating the prediction of metal-binding proteins. Our software can be accessed at the SVMProt server

    Unique Carboniferous-Permian tectonic-metallogenic framework of Northern Xinjiang (NW China): Constraints for the tectonics of the southern Paleoasian Domain

    The late Paleozoic tectonic-metallogenic framework of North Xinjiang of the southern Paleoasian Domain was characterized by a serious of Carboniferous-Permian events, including: (1) late Carboniferous-Permian Chinese Altay island arc and its metamorphism, granulite in the Chinese Altay, radiolarian chert and high-pressure/ultra-high-pressure metamorphism; (2) late Carboniferous-early Permian adakites, Alaskan-type mafic-ultramafic complexes, and calc-alkaline magmatism, together with porphyry copper deposits, which occurred in the North Xinjiang; and (3) late Carboniferous ophiolite and island arc volcanic rocks located in the Tian Shan. Combined with the facts that there was no typical foreland basin, no typical collisional-type granitoid, and there were large amount of strike-slip faulting, it is suggested that in the Carboniferous-early Permian North Xinjiang was characterized by the coexistence of compression-extension-strike-slip structures with active magmatism and metallogeny. These phenomena all indicate that there were active margins during the late Carboniferous-early Permian, leading to the notion that the complicated accretionary orogeny along the southern Paleoasian Domain may have lasted to the latest Carboniferous-Permian.新疆北部晚古生代独特的构造一成矿作用以发育大量石炭纪一二叠纪构造一成矿事件为特征, 其中包括: (l) 发育于晚石炭世一二叠世的阿尔泰岛弧及其变质事件、阿尔泰麻粒岩与基性杂岩、西南天山放射虫硅质岩和高压一超高压一低压麻拉岩相变质事件; (2 )北疆发育的石炭纪(一二叠世)埃达克岩一高镁安山岩一富N d 玄武质岩组合、阿拉斯加型基性一超基性杂岩和大量的与俯冲相关的钙碱性岩浆活动与斑岩型铜矿床成矿作用; (3) 天山晚石炭世晚期蛇绿岩与岛弧火山岩等。结合北疆地区相关的前陆盆地发育不明显、碰撞型花岗岩欠发育与大量发育平行造山带大型走滑构造等现象, 可以认为新疆北部在石炭纪一二叠纪挤压一伸展一走滑并存, 岩浆活动与成矿作用活跃。这些新进展表明新疆北部在晚石炭世一二叠纪可能仍存在活动陆缘, 因此, 古亚洲洋构造域南部复杂增生造山作用最后延至晚石炭世晚期一二叠纪。published_or_final_versio

    Expression, purification, and characterization of a novel Ca2+- and phospholipid-binding protein annexin B2

    Annexin B2 (AnxB2) is a novel member of the annexin family of Ca2+- and phospholipid-binding proteins from Cysticercus cellulosae. To obtain highly pure AnxB2 with an easy and inexpensive purification approach, its cDNA was cloned into the prokaryotic expression vector pJLA503 and the translation initiation codon was immediately under the control of the inducible bacteriophage λ promoters PR and PL. After induction by shifting temperature, large amounts of non-fusion protein were produced in Escherichia coli in a soluble form. Then a novel purification method based on Ca2+-dependent phosphatidylserine (PS)-binding activity was established, whereby the purity of AnxB2 was increased to 98.7%. Western blot analysis showed that recombinant AnxB2 was specifically recognized by serum of pigs infected with cysticercosis. In vitro test showed that, the recombinant AnxB2 had anticoagulant activity and platelet binding activity.     Trajectories of self-rated health in people with diabetes: Associations with functioning in a prospective community sample

Relatively little is known about changes and the trajectories of SRH in people with chronic medical conditions. The aims of the present study were to identify and describe longitudinal trajectories of self-rated health (SRH) status in people with diabetes. Methods: A prospective community study was carried out between 2008 and 2011. SRH was assessed at baseline and yearly at follow-ups (n=1288). Analysis was carried out through trajectory modeling. The trajectory groups were subsequently compared at 4 years follow-up with respect to functioning. Results: Four distinct trajectories of SRH were identified: 1) 72.2% of the participants were assigned to a persistently good SRH trajectory; 2) 10.1% were assigned to a persistently poor SRH trajectory; 3) mean SRH scores changed from good to poor for one group (7.3%); while 4) mean SRH scores changed from poor to medium/good for another group (10.4%). Those with a persistently poor perception of health status were at higher risk for poor functioning at 4 years follow-up than those whose SRH scores decreased from good to poor. Conclusions: SRH is an important predictor for poor functioning in diabetes, but the trajectory of SRH seems to be even more important.     Top Quarks as a Window to String Resonances

    We study the discovery potential of string resonances decaying to ttˉt\bar{t} final state at the LHC. We point out that top quark pair production is a promising and an advantageous channel for studying such resonances, due to their low Standard Model background and unique kinematics. We study the invariant mass distribution and angular dependence of the top pair production cross section via exchanges of string resonances. The mass ratios of these resonances and the unusual angular distribution may help identify their fundamental properties and distinguish them from other new physics. We find that string resonances for a string scale below 4 TeV can be detected via the ttˉt\bar{t} channel, either from reconstructing the ttˉt\bar{t} semi-leptonic decay or recent techniques in identifying highly boosted tops.Comment: 22 pages, 6 figure

    Distinct Neurocognitive Strategies for Comprehensions of Human and Artificial Intelligence

    Although humans have inevitably interacted with both human and artificial intelligence in real life situations, it is unknown whether the human brain engages homologous neurocognitive strategies to cope with both forms of intelligence. To investigate this, we scanned subjects, using functional MRI, while they inferred the reasoning processes conducted by human agents or by computers. We found that the inference of reasoning processes conducted by human agents but not by computers induced increased activity in the precuneus but decreased activity in the ventral medial prefrontal cortex and enhanced functional connectivity between the two brain areas. The findings provide evidence for distinct neurocognitive strategies of taking others' perspective and inhibiting the process referenced to the self that are specific to the comprehension of human intelligence

    Chronic hepatitis B: whom to treat and for how long? Propositions, challenges, and future directions

    Recent guidelines of the American Association for the Study of Liver Diseases, the European Association for the Study of the Liver, and the Asian Pacific Association for the Study of the Liver 2008 update of the “Asian-Pacific consensus statement on the management of chronic hepatitis B” offer comprehensive recommendations for the general management of chronic hepatitis B (CHB). These recommendations highlight preferred approaches to the prevention, diagnosis, and treatment of CHB. Nonetheless, the results of recent studies have led to an improved understanding of the disease and a belief that current recommendations on specific therapeutic considerations, including CHB treatment initiation and cessation criteria, particularly in patient populations with special circumstances, can be improved. Twelve experts from the Asia-Pacific region formed the Asia-Pacific Panel Recommendations for the Optimal Management of Chronic Hepatitis B (APPROACH) Working Group to review, challenge, and assess relevant new data and inform future updates of CHB treatment guidelines. The significance of and controversy about reported findings were discussed and debated in an expert meeting of the Working Group in Beijing, China, in November 2008. This review paper attempts to identify areas requiring improved CHB management and provide suggestions for future guideline updates, with special emphasis on treatment initiation and duration

    First observation of psi(2S)-->K_S K_L

    The decay psi(2S)-->K_S K_L is observed for the first time using psi(2S) data collected with the Beijing Spectrometer (BESII) at the Beijing Electron Positron Collider (BEPC); the branching ratio is determined to be B(psi(2S)-->K_S K_L) = (5.24\pm 0.47 \pm 0.48)\times 10^{-5}. Compared with J/psi-->K_S K_L, the psi(2S) branching ratio is enhanced relative to the prediction of the perturbative QCD ``12%'' rule. The result, together with the branching ratios of psi(2S) decays to other pseudoscalar meson pairs (\pi^+\pi^- and K^+K^-), is used to investigate the relative phase between the three-gluon and the one-photon annihilation amplitudes of psi(2S) decays.Comment: 5 pages, 4 figures, 2 tables, submitted to Phys.
